Metabolism

After ingesting Fenbendazole, the drug is transformed through two processes in the body namely Oxidation and hydroxylation.

 

The first process takes place with the aid of liver enzymes – CYP2J2 and CTP2C19. This process hydroxylates Fenbendazole to create a new metabolite; hydroxytenbendazole.

 

The second process is the oxidation of the sulfide group. This is catalyzed in the liver by the enzymes CYP3A and flavin-containing monooxygenase. As a result, a new compound forms – oxfendazole (which also displays anticancer and anthelmintic properties)

In the second process, the sulfide group undergoes oxidation. The enzyme CYP3A and flavin-containing monooxygenasecatalyze the oxidatedsulfide group in the liver. At the end of the catalyzations, a new compound is formed; Oxfendazole (which has been found to possess anticancer and anthelmintic characteristics.

This process interestingly reveals that Fenbendazole is a drug and a prodrug at the same time considering how it was able to create another active medication.

Due to the bioavailability of Fenbendazole powder, it is not completely converted to its metabolites. We are only able to absorb a small number of Fenben via the gastrointestinal tract. Eventually, Fenbendazole and others like Oxfendazole and hydro-fenbendazole are passed out through the anus.

Note that one of the enzymes involved in metabolizing Fenbendazole CTP2C19 is the same enzyme involved in the metabolism of such drugs asastemizole,mesoridazine, apixaban, tamoxifen, ebasite, cyclosporine, and thioridazine
